brief introduction
colifast online monitor - calm is a fully automatic early warning system for monitoring coliform bacteria, Escherichia coli, heat-resistant Escherichia coli and Pseudomonas aeruginosa in water. The detection time ranged from 2 to 12 hours. This technology is time-saving, low-cost and environmentally friendly. Calms have many mature cases in monitoring raw water, water being treated, wastewater and reclaimed water. No laboratory equipment or microbiological background is required to operate this instrument. Colifast calm can connect with system computer or LAN port. Colifast culture medium is patented and can be provided in the pre filled multi well disk. The system is flexible and can adapt to different applications. Calms can be used for parallel analysis of different strains. According to the method and test evaluation rate, calm needs to change reagents monthly or weekly.
method principle
cal instrument can detect and quantify E.coli by fluorescence monitoring of b-glucuronidase activity (E.coli) or p-galactosidase activity (total coliform and thermotolerant coliform). The detection of Pseudomonas aeruginosa is based on substrate hydrolysis of aminopeptidase. Colifast technology is a combination of unique media formula and instrument detection system. This technology has been patented in many countries and is applying for several international patents. The growth of other bacteria was prevented by the suppressant in the medium and the sample culture at high temperature: the culture temperature of heat-resistant coliform group was 44 ℃, and that of total coliform group / Escherichia coli was 37 ℃.
two measurement methods are available
one of the two methods for measuring the instrument at one time. The most probable number (MPN) method is based on bacterial culture. Adding a certain volume of 4-6 well growth medium, the number of bacteria depends on the number of wells with bacteria growth after the incubation period (11-12 hours). Mixed sampling can make the results more accurate.
rapid measurement is also a quantitative method, based on the enzyme activity of bacteria in water samples. It can respond in 1-2 hours, so it is very suitable for water sources that may have major pollution events.
calm is a flexible instrument. This means that the instrument can adapt to different applications and different bacterial concentrations. If the average concentration of bacteria reaches 500 CFU / 100mL, the system will automatically sample 0.2ml for positive test. By injecting about half of the volume of water sample (about 0.1ml), both positive and negative water samples can be obtained at the same time. If the concentration of bacteria is low, 10ml of water sample can be sampled to change the concentration of culture medium.
